The cryopreservation of canine semen can be performed utilizing either
the small 0.5ml straws (desireable when the semen will be used for
surgical or transcervical insemination by direct injection into the
uterus), or the larger 5ml "Macrotubes" we commonly use for equine semen
(preferable for vaginal AI). A minimum dose is generally considered to
be 100 million, progressively motile cells, whether the semen is fresh
or frozen-thawed. With frozen semen, therefore, we usually aim for about
400 million total cells in a dose to begin with. At a "minimum" postthaw
motility of 30%, this will give us 120 million motile cells for the
resulting
insemination.
Dogs produce between one and about 15 straws per collection,
depending upon age, breed, and health status. Smaller dogs produce less
than larger dogs, and as with any species, younger adults produce the
best quality samples. We normally test four different extenders at
first, one of which we have developed ourselves. This "test-freeze"
determines which extender we use in subsequent freezing sessions.
For most males, collection once per week provides optimal results for
both semen quality and quantity. While a teaser female is rarely
required, we seem to obtain more sperm when one happens to be "handy".
Any surgical insemination work is performed a few miles away
in a colleague's surgical suite, although this method is being rapidly
replaced by the transcervical procedure using endoscopic visualization
of the cervix, which we perform and recommend. It is quicker, less
expensive, less risky, and less stressful, with similar pregnancy rates.
We have also developed a shipping extender for fresh-chilled semen
that maintains exceptional longevity; in some cases over several weeks!
Because we make these extenders ourselves, from the very finest
chemicals available, and are not
required to use proprietary media sold other networks, we can offer semen
cryopreservation to the breeder at an extremely competitive fee.
We have been inspected by the AKC and are officially approved by them
as a semen collection center.
